The mail client in Windows 10 Mobile expects the tiles to have specific ids for the notifications to work - if you tap on the email account, bring up the menu via the ... button at the bottom of the screen, you'll see that it doesn't think the folder is pinned. Unfortunately you can't. This feature used to be in some of the preview builds but Microsoft pulled it out, citing issues with Start screen layouts. They said it would be quite some time before it was put back in, and given the current rate of development I wouldn't expect to see it any time soon, if ever.
